ABSTRACT

Objective@#To evaluate the risk factors of postoperative upgrade to pT3a of cT1 renal cell carcinoma, and to establish a nomogram prediction model to improve the ability of predicting locally advanced renal cell carcinoma and provide a reference for clinical surgical decision-making.@*Methods@#Clinical data of 1 376 patients with cT1 (diameter ≤ 7 cm) renal tumor hospitalized for surgery from January 2010 to December 2016 were retrospectively analyzed. There were 979 males and 397 females, with the mean age of (57.65±10.92) years. The mean body mass index (BMI) was (25.47±3.27) kg/m2 and the average tumor size was (4.02±1.52) cm. There were 711 tumors on the left and 665 on the right. There were 363 cases with clinical symptoms, 567 patients with smoking history , 732 cases with history of chronic disease. There were 289 cases with tumor necrosis, 636 cases with tumor protrusion, 822 cases with irregular tumor , and 738 cases with renal sinus compression. Partial nephrectomy and radical nephrectomy were performed in 396 cases and 980 cases respectively. Mann-whitney U test and chi-square test were used for univariate analysis, logistic regression analysis was used for multivariate analysis to analyze the predictors of upgrading, R software was used to construct the nomogram predictive model, C-index was used to evaluate the model discrimination, and calibration curve method was used to evaluate the consistency of the model.@*Results@#Postoperative pathology of total 1 376 cases showed that there were 1 195 cases of clear cell carcinoma of kidney, 48 cases of papillary cell carcinoma, 57 cases of chromophobe cell carcinoma, and 76 cases of other types. Among the 1 376 patients with cT1 renal tumor, 75 patients were upgraded to pT3a, accounting for 5.5% of all patients. Univariate analysis showed that the patients who upgraded to pT3a were older [(63.08±10.17) years old and (57.34±10.88) years old], and the tumor length and diameter were larger [(5.24±1.35) cm and (3.95±1.51) cm]. Patients with clinical symptoms [46.7% (35/75) vs. 25.2%(328/1 301)], patients with CT indication of tumor necrosis [40.0%(30/25 975) vs. 19.9% (259/1 301)], patients with irregular tumor contour [73.3%(55/76 775) vs. 59.0%(767/1 301)], and patients with radical nephrectomy were higher [(92.1% (70/91 075) vs. 70%(910/1 301)]. All the differences were statistically significant (P<0.01). Multivariate analysis showed that the independent predictors of upgrading were age (OR=1.046, P<0.001), larger tumor (OR=1.504, P<0.001), clinically symptom (OR=2.153, P=0.004), irregular tumor profile (OR=2.466, P=0.002), and tumor necrosis on CT (OR=2.588, P<0.001). The C-index was 0.808, the calibration curve of forecasting curve with the standard curve fit was good, and the prediction of renal cancer are better in predict consistency.@*Conclusions@#Based on the five preoperative predictors, including age, tumor size, clinical presence or absence of symptoms, tumor profile, and whether or not the tumor necrosis indicated by CT, this study developed a nomogram of cT1 renal cancer upgrade to pT3a. This nomogram has a good statistical significance, and this model can provide prognosis consultation for patients and provide reference for doctors to make decisions before treatment.

ABSTRACT

Objective To explore the expressions and clinical significance of matrix metalloproteinase9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in renal carcinoma.Methods A total of 45 patients with renal carcinoma received treatment at our hospital from January 2015 to January 2017 were enrolled as observation group,and 40 healthy subjects in the same period were enrolled as control group.The expressions of MMP-9 and TIMP-1 in observation group and control group were tested and compared by enzymelinked immunosorbent assay.The relationship between MMP-9 and TIMP-1 and their correlations with the clinical pathological features were analyzed.Results The serum levels of MMP-9 in observation group were higher than those in control group,and the differences were significantly significant [(461.7 ± 53.6) pg/ml vs.(142.9±31.7) pg/ml;t=32.840,P<0.001],TIMP-1 [(387.9±11.8) pg/ml vs.(136.5 ±21.8) pg/ml;t=13.290,P<0.001] andMMP-9/TIMP-1 [(1.2±0.2) vs.(1.1 ±0.1);t=3.640,P<0.001].Correlation analysis showed that the serum expression level of MMP-9 had significantly positive correlation with TIMP-1 for patients in observation group (r =0.847,P < 0.001).The serum expression levels of MMP-9 and TIMP-1 for patients in observation group were related to the lymph node state (t =2.657,P =0.011;t =4.420,P <0.001) and primary tumor staging (t =6.200,P <0.001;t =5.042,P <0.001),but they were not related to pathological type (t =1.130,P =0.265;t =0.109,P =0.914).Conclusion The peripheral blood levels of MMP-9 and TIMP-1 are increased in patients with renal carcinoma,and they are associated with the progression of disease,which can be used to evaluate the progress of disease.

ABSTRACT

Objective@#To detect the presence of HPV associated antibodies in the serum of patients diagnosed with esophageal cancer in Chaoshan region and to provide data that could possibly be used as reference for the monitoring of patients with esophageal cancer.@*Methods@#The pSecTag recombinant vectors containing the HPV16 E6/E7 and HPV18 E6/E7 genes were constructed respectively to express HPV16 E6/E7 and HPV18 E6/E7 fusion proteins in 293 cell line after transfecting the cell line. Immunoenzymatic method was employed with the fusion proteins as antigen to detect IgG antibody against HPV in serum of 76 esophageal cancer patients and 149 normal persons undergoing health checkup. In addition, the same sera were assessed by enzyme-linked immunosorbent assay (ELISA) method using L1 as antigen.@*Results@#Only one esophageal cancer patient’s serum presented as positive for IgG by immunoenzyme technique when E6/E7 was used as antigen. However, when the L1 was used as antigen in ELISA assay, 37 of the 76 cases of esophageal cancer patients (48.7%) were HPV antibody-positive. Among the 149 health checkup persons, 79 (53.0%) were HPV antibody-positive. No significant difference was found between the two groups (P>0.05).@*Conclusion@#HPV related antibodies in the serum remain inapplicable as a screening tool for serological detection of esophageal cancer. Further research is needed to understand the role and diagnostic applications of HPV infection in esophageal cancer.

ABSTRACT

BACKGROUND:Tropomyosin 4 level has been found to be an increase in the spinal cord based on the 2-DE/MALDI-TOF/MS method. However, there is little report about the relationship between tropomyosin 4 and pathogenesis and progress of spinal cord injuries. METHODS/DESIGN:Randomized control ed trial:rat models of complete spinal cord transection were made and expression levels of tropomyosin 4 at 3-28 days after modeling were determined by two-dimensional electrophoresis, animo acid serie analysis, quantitative PCR and western blot. Experiment for exporing the genetic mechanism:effects of tropomyosin 4 scilencing by lentivirus recomnination technology on the dendrite length of spinal cord neurons in vitro were observed, and its effects on the neurological function of rats after complete spinal cord transaction were assessed through Basso, Beattie, and Bresnahan scoring. DISCUSSION:This study wil be powered to provide a novel and effective treatment strategy for neurological function recovery after spinal cord transection based on the lentivirus recomnination carrying tropomyosin 4, as wel as optimistic future for clinical gene treatment of complete spinal cord transaction through figuring out the underlying mechanism. ETHICAL APPROVAL:This study was approved by the Ethics Committee of Kunming Medical University, China. The surgical operation and postoperative care of rats were in line with the rules of Chinese Experimental Animal Protection and Ethics Committee, and the guideline of the National Institutes of Health

ABSTRACT

Objective To investigate the expression levels of BDNF, trkB and ChAT mRNA and proteins in the brain of adult tree shrews ( Tupaia belangeri ) .Methods Quantitative real-time PCR was employed to detect the expression levels of BDNF, trkB and ChAT mRNA in the hippocampus, basal ganglia and frontal cortex of adult tree shrews.The expression levels of BDNF, trkB and ChAT proteins andβ-actin was used as internal standard.Results The expression level of BDNF mRNA was highest in the hippocampus of adult tree shrew, and there were significant differences between that in the hippocampus, and basal ganglia and frontal cortex (P0.05) in the expressions of trkB protein among the hippocampus, basal ganglia and frontal cortex of the adult tree shrews.There were no significant differences in expressions of ChAT mRNA and protein among the hippocampus, basal ganglia and frontal cortex in adult tree shrews ( P>0.05 ) . Conclusions The expression levels of ChAT mRNA were consistent with that of ChAT protein in the hippocampus, basal ganglia and frontal cortex of adult tree shrews, while the expression levels of BDNF and trkB mRNA were not consistent with their proteins, which might indicate that the transcriptional regulation pattern might be more complex.Tree shrew is a valuable animal model in the study of mechanism of BDNF/trkB gene expression.

ABSTRACT

We wished to screen the cell line that stably expresses the HPV18E5 protein, and to ascertain the influence of HPV18E5 protein on cell proliferation and the cell cycle. The HPV18E5 gene was amplified by the polymerase chain reaction. Then, the His-tag pSecTag-HPV18E5 eukaryotic expression vector was constructed by digestion ligation and connection. The recombinant plasmid was transfected into Balb/c3T3 cells with lipofectamine, and positive cell lines were screened by a culture medium containing bleomycin. HPV18E5 expression in cells was confirmed by western blotting and immuno-enzymatic methods. The influence of HPV18E5 on cell proliferation and the cell cycle were detected by Cell Counting Kit-8 and flow cytometry, respectively. The pSecTag-HPV18E5 eukaryotic expression vector was constructed. After 21-day selection in a culture medium containing 400 μg/mL bleomycin, stably expressing HPV18E5 protein cells were harvested. Compared with control groups, cell proliferation in HPV18E5 stably expressed cells was obviously increased, as was the S phase in the cell cycle. Our results suggested that HPV18E5 influences cell proliferation and the cell cycle. Our study has laid the foundation of the biologic properties of HPV18E5 protein, which will aid further studies on the mechanism of action of carcinogenesis.

ABSTRACT

Objective To investigate the hand hygiene status of urban and rural residents in Hangzhou City of Zhejiang Province to provide scientific base for health promotion.Methods Urban residents,rural residents,personnel in government institutions,urban students,rural students and enterprise workers were enrolled in this investigation.By using constant volume sampling method,fifty individuals in each group were recruited from 13 districts of Hangzhou City,and a total of 3 337 people filled in the questionnaire,which included demographic characteristics,knowledge of hand hygiene and hand washing.Logistic regression analysis and x2 test were used for data analysis.Results Correct hand-washing was found in 35.84% urban and rural residents (urban vs.rural:41.45% vs.27.06%).Awareness rate of hand hygiene was 35.06%.The influencing factors of proper hand washing were location,gender,age,educational level and understanding of hand hygiene.The influencing factors of awareness of hand hygiene were location,gender,age and educational level.Conclusion Hangzhou citizens had a relatively lower rate of proper hand washing,and significant difference was found between urban and rural residents; the study suggested that para-urben residents need to improve their knowledge and behavior of hand hygiene.Meanwhile,the students urgenty need to improve their hand hygiene

ABSTRACT

Objective To analyze and compare the incidence of diarrhea between cerebral hemorrhage and infarction. Method We observed and compared the time when diarrhea occurred in both 147 cases of cerebral hemorrhage patients and 142 cases of cerebral infarction patients by using statistical methods. Result The incidence of diarrhea in former group was 36.05%, and that of latter was 16.91%. Conclusions The incidence in patients with diarrhea of cerebral hemorrhage was markedly higher than that of patients with cerebral infarction. This may attribute to the higher intracranial pressure in cerebral hemorrhage patients than that of cerebral infarction ones. The complications of reduction of immune function,intestinal infection and hypoproteinemia may be the other factors resulting in the higher incidence of diarrhea in cerebral hemorrhage patients than that of cerebral infarction ones.

ABSTRACT

Objective To lay the physiological and biochemical basis for establishing and evaluating tree shrews model of human disease. Methods There were 92 Tupaia belangeri chinensis, in which half of them were male,they were allowed to eat nothing for 12 hours, then we sampled heart blood 0.8~1.0 mL without anesthesia and put blood samples into sterilized centrifuge tube for separation and preparation of serum. Olympus AU5400 automatic biochemistry analyzer was used to measure biochemical indexes. Then 1.5~2.0 mL of urine of each tree shrew was collected and put into sterilized centrifuge tube for measuring renal function by using Combi-scan500 urine analyzer. Fianally SPSS statistics software was used to analyse the measured values, and comapared the measured values with the human reference values. Results There were significant differences in myocardial enzymes and some renal function indexes such as lactic dehydrogenase,α- hydroxybutyric, acid dehydrogenase, creatinine, uric acid, urine specific gravity and pH value between male and female Tupaia belangeri chinensis ( 0.05) . Then determination value of Tupaia belangeri chinensis, male’s and female’s, myocardial enzyme and part of the renal function indexes were compared with the human reference values. Some indexes including urea, urine specific gravity, urine, urobilinogen were in the range of human reference value, while the values of urine bilirubin, urine nitrite. lactate dehydrogenase, creatine kinase, α-hydroxybutyrate dehydrogenase, creatine kinase isoenzyme CKMB were higher than the human reference value. White blood cell, urine protein, ketone, occult blood most of them were negative as the same to human reference value, but sometimes were positive, and the positive rates respectively were 3.95%and 46.1%,39.5%,2.63%. The measured value of Vitamin C was positive that is completely opposite to human reference value, but sometimes is negative, the negative rate was 6.6%. Conclusion Urea, urine specific gravity, urine, urobilinogen, urine bilirubin, urine nitrite can be directly used as the indexes for evaluating tree shrews models of human disease, other indexes can be used as indexes for judgment of the normal physiological and biochemical basis of tree shrews.

ABSTRACT

Objective To summary the experience of microsurgery of meningioma in skull base treatment. Methods To study the clinical data of 10 cases patients of meningioma in skull base who were subjected to microsurgery by using retrospective analysis. Among the 10 patients, there were 3 cases of meningioma of the olfactory groove. There were 2 cases of sphenoid ridge meningioma, meningioma in the middle cranial fossa, meningioma in the cerebellopontine angle respectively.There was 1 case of meningioma of velarium in temporal lobe. Results In 7 cases, the meningioma was completely removed. And in another 3 cases, the meningioma got subtotal resection. During and after the operation, there were no severe complications observed or death occurrence in all the 10 cases of meningioma patients. Two months post of operation,all patients got a significant amelioration in their symptoms. A total of 9 cases were followed up. There was 1 case of IIa type meningioma recurred. Conclusion To elevate the whole shearing rate of meningioma in skull base and reduce the mortality of it,it is most important for appropriate operative route selection and well mircosugical technique application.

ABSTRACT

Objective To investigate the effect of gastrodin on brain-derived neurotrophic factor (BDNF) expression in Alzheimer's disease ( AD) tree shrew. Methods AD tree shrew model was established by intracerebroventricular injection with amyloid-βpeptide (Aβ) . Since the eighth day after injection, the tree shrews were treated with gastrodin for thirty days. The expression of BDNF in tree threw hippocampus was detected by reverse transcription polymerase chain reaction (RT-PCR) . Results The expression of BDNF mRNA in the treatment group was higher than that in the model group ( <0.05) . Conclusion Gastrodin can increase some degree of the expression of BDNF.

Objective:To predict and analyze the secondary structure and B cell epitopes of Izumo protein.Methods: The secondary structure and flexible regions of Izumo protein were predicted by the methods of Chou-Fasman,Gamier-Robson and Karplus-Schulz.Moreover,hydrophilicity plot,surface probability plot and antigenic index of Izumo protein were predicted by the methods of Kyte-Doolitde,Emini and Jameson-Wolf,respectively.Results: Izumo protein contained moreαhelix regions.There were several centers ofαhelix in the regions of 6-17,30-40,88-99,103-120,153-160,173-188,249-260,283-297,334-338 and 339-346 of Izumo protein,and several centers of βsheet in the regions of 21-25,198-200,245-248 and 320-323.Moreover,many distinct B cell epitopes in Izumo protein possibly localized in the regions of 3642,62-66,94-99,118-122,129-132,151-154,161-164,173-177,205-208,212-216,256-265,271-276,283-288,314-318 and 336-350.Conclusion:These results are helpful for identification of the dominant B cell epitopes and the functional domains of Izumo protein.

ABSTRACT

EV71 infection has become a serious public health threat especially among young children. Yet, at present, no specific antiviral drug against EV71 infection is available. A number of scientists are studying various kinds of vaccines, including inactivated vaccine, virus-like particle vaccine, DNA vaccine, synthetic peptide vaccines, and transgenic oral vaccine. This article reviews the recent advancement in the design of various kinds of vaccine against EV71 as well as their prospective usefulness, effectiveness, weakness and developments in the foreground.

The present study investigated different types of eoexpression of brain derived neumtmphic factor(BDNF),nerve growth factor(NGF)and neutmphin-3(NT-3)mRNA and/or proteins in the left sixth lumbar dorsal root ganglion(DRG)of cats and discuss themechanism of coexpression in order to provide foundation for elucidating the relationship between the expression of neurotrophic factors andspinal cord plasticity.The eats used in this study were normal animals without any interventional treatment.They were subjected to renloveof the left L6 DRG and their DRG were processed for immunohistechemistry and in situ hybridization double staining to observe whetherthere are coexpression of mRNA and proteins of BDNF,NGF and NT-3.The results showed that the pmteios and mRNA of BDNF,NGFand NT-3 were all expressed in the DRG of cats,but the types of coexpression of mRNA and proteins were different and diverse amongthese three neumtrophic factors.The results of immunohiatochemistry showed that BDNF immunoreactivities were mainly observed in thecytoplasm and nucleus,and the staining of nucleus was weaker than that of cytoplasm;NGF immunoreactivities were mainly observed innucleus while NT-3 mainly in cytoplasm.The results of in situ hybridization showed that BDNF and NGF positive signals mostly distributedin the cytoplasm,NT-3 positive signals were observed in both the cytoplasm and nucleus.Our results suggest that the proteins and mRNAof BDNF,NGF and NT-3 have different types of coexpression which indicate they may have autocrine and/or paracrine mechanism contrib-uting to the plasticity of spinal cord in the left L6 DRG of cats.

Objective To establish a serial of stable and mature methods of primary culture hippocampus neurons of GFP-transgene embryonic mice,to get the morphologic data of cultured neurons.Based on this research,in future,we can give an important theoretical and practical support for the therapy of nervous system diseases by transplanting with hippocampus neurons of GFP-transgene embryonic mice.Methods We primarily cultured the hippocampus neurons derived from the GFP-transgene embryonic mice in vitro.Under the microscope,we found cultured hippocampus neurons could live for more than one month and appeared to be the best status in 5~7 d after culture.During this time,the processes of the neurons are thick and the neurons connected one another to form the"cells-net" through their processes.After 14 d,the growth of the hippocampus neurons became slow.Results A serial of culture methods of hippocampus neurons had been successfully established.These cultured neurons were identified by the immuno-histochemical methods.They grow well in different phases before 14 d after culture.Conclusion Culturing hippocampus neurons of GFP-transgene embryonic mice is a simple,stable and effective method which can be applied to scientific research by other researchers.

ABSTRACT

Objective To study the function of cyclooxygenase 2(COX 2) and its specific inhibitor NS 398 on the cell growth and invasion ability of urothelial carcinoma cell line EJ. Methods The cox 2 cDNA was transfected into the urothelial carcinoma cell line EJ and a cell line EJ COX 2 which highly expressed cox 2 gene permanently was gained. The cell growth rate before and after transfection was observed. Then at various concentrations of NS 398, the invasion ability was detected by Boyden Chamber and expression levels of uPA by RT PCR and Western blot. Results The EJ COX 2 cell line grew more rapidly and had a stronger invasion ability than EJ and its uPA expression increased significantly. NS 398 could dose dependently inhibit the expressions of COX 2 and uPA and the invasiveness of EJ COX 2 cell. Conclusion COX 2 can stimulate the growth of urothelial cell line EJ and promote its invasion ability by stimulating the expression of uPA.

ABSTRACT

Objective To investigate the effect of thymosin ?1 (T?1) on cellular immune function in gastric cancer patients through observing its treatment on the differentiation of T-lymphocyte subsets from screened peripheral blood mononuclear cells (PBMCs). Methods PBMCs were obtained by centrifugation of blood samples from 18 healthy subjects and 32 patients with gastric cancer,and then cultured in the presence of culture medium with addition of T?1 at 50,10 and 1 ?g/ml for 2 d. T lymphocyte subsets (such as CD4+,CD8+ and CD4+ CD25+ Foxp3+ T cells) and Th1/Th2 multiplex cytokines were detected by flow cytometry (FCM). Results After PBMCs isolated from healthy people and patients were incubated with or without T?1,there was no significant change in percentage of CD4+,CD8+ peripheral lymphocyte subsets and ratio of CD4+/CD8+. There was no obvious change in the percentage of CD4+ CD25+ Foxp3+ T lymphocyte subsets in the normal control,but a significant increase was observed in the cells from patients with gastric cancer after treatment (P

ABSTRACT

Objective To explore the role of Bcl-2 and Caspase-3 in modulating apoptosis of ER-negative breast cancer cells induced by tamoxifen. Methods ER-negative breast cancer cell lines MDA-MB-231 were treated with 10.0?M tamoxifen for 12, 24, 36,48, 60 hours. The rate of cell apoptosis with or without caspase-3 inhibitor Ac-DEVD-CHO, and protein expression of Bcl-2,Bax were determined by flow cytometry, and the activity of Caspase-3 was examined with fluorophotometry. Results The expression of Bcl-2 was down-regulated, the activity of Caspase-3 and the rate of cell apoptosis were increased by TAM time-dependently, and the rate of apoptosis reached its peak at 48 hours. The expression of Bcl-2 was (negatively) correlated with activity of caspase-3. Tamoxifen, however, did not affect Bax protein expression. Ac-DEVD-CHO, a caspase-3 inhibitor, blocked the activation of caspase-3 and inhibited cell apoptosis (induced) by tamoxifen. Conclusions TAM could induce apoptosis in ER-negative breast cancer cells via (mitochondria) pathway by down-regulating Bcl-2 expression, and the activation of Caspase-3 might play an (important) role in the process of tamoxifen-induced apoptosis of ER-negative breast cancer cells.
